Gene expression profiling indicates a shift in ammonia assimilation capacity along the hepatic acinus induced by different forms of selenium in vitamin–mineral mixes fed to beef steers grazing on toxic endophyte-infected tall fescue

چکیده

Recently, we demonstrated that supplementing (with 3 mg Se per day) the diets of growing beef steers grazing on Se-deficient toxic endophyte-infected tall fescue-based forage with either organic (OSe, SEL-PLEX) or a 1:1 blend (MIX) OSe and inorganic (ISe, sodium selenite) in vitamin–mineral mixes, rather than forms Se, ameliorated several classic serum symptoms fescue toxicosis. Importantly, higher levels hepatic glutamine synthetase activity were observed MIX steers. Accordingly, transcriptome level targeted mRNA expression analyses conducted same liver tissue to determine if treatments affected other metabolic pathways, especially those are responsible for supplying substrates synthetase. The effect ISe, OSe, ( n = 8/treatment) relative abundances [determined using microarray real-time reverse-transcription PCR (RT-PCR)] protein (determined Western blotting) was assessed by ANOVA. Fisher’s protected LSD procedure used separate treatment means, significance being declared at p ≤ 0.05. Microarray analysis identified < 0.01, false discovery rate of< 33%) 573 annotated differentially expressed gene (DEG) transcripts. Canonical pathway DEGs central glutamate biosynthesis/degradation proline biosynthesis. Targeted RT-PCR found had lower periportal ammonia-assimilation urea-synthesizing capacities (lower glutaminase 2, key ornithine cycle enzymes, mitochondrial ornithine/citrulline exchanger mRNA) ISe In addition, capacity pericentral ammonia assimilation (higher activity) production hepatocytes from α-ketoglutarate dehydrogenase, decreased arginase 2 aminotransferase mRNA). form supplemental also steers’ metabolism, having synthesis pyroline-5-carboxylate synthesis. conclusion, shifts urea production, thus potentially increasing whole-animal N efficiency supply hepatic-synthesized glutamine.

Postgraze assessment of toxicosis symptoms for steers grazed on toxic endophyte-infected tall fescue pasture.

A 2-yr pen experiment was conducted using 12 different crossbred Angus steers each year to determine if short-term changes in prolactin concentrations, body temperature, and vasoconstriction reflect recovery from fescue toxicosis after steers that previously grazed toxic endophyte (Neotyphodium coenophialum)-infected Kentucky 31 tall fescue [Lolium arundinaceum (Schreb.) Voluntary intake and ingestive behavior of steers grazing Johnstone or endophyte-infected Kentucky-31 tall fescue.

Effects of grazing low-endophyte (Acremonium coenophialum Morgan-Jones and Gams, less than 1% infection) Johnstone (J) or high-endophyte (60% infection) Kentucky-31 (K) tall fescue (Festuca arundinacea Schreb.) on grazing behavior and voluntary intake were studied. Six Angus steers (average initial BW = 326 kg) grazed 1.21-ha plots of each forage cultivar (three steers per cultivar) in four 28-...

Pituitary genomic expression profiles of steers are altered by grazing of high vs. low endophyte-infected tall fescue forages

Consumption of ergot alkaloid-containing tall fescue grass impairs several metabolic, vascular, growth, and reproductive processes in cattle, collectively producing a clinical condition known as "fescue toxicosis." Despite the apparent association between pituitary function and these physiological parameters, including depressed serum prolactin; no reports describe the effect of fescue toxicosi...

Novel endophyte-infected tall fescue for growing beef cattle.

Tall fescue (Festuca arundinacea, Shreb.) is the predominant cool-season, perennial grass in the eastern half of the United States, and the majority is infected with the endemic endophyte (E+) Neotyphodium coenophialum, resulting in millions of dollars in revenues lost to the beef industry. Endophyte-free (E-) tall fescue was initially tapped as a "silver bullet" for the solution to fescue toxi...

Novel Endophyte-Infected Tall Fescue

Tall fescue (Schedonorus phoenix (Scop.) Holub) is a cool season perennial grass that fills a critical forage produc-tion gap in the Southeast and is the predominant forage crop in much of the U.S. In fact, this species is the most widely used grass in improved U.S. pastures, occupying more than 36 million acres. Its popularity is related to establishment ease, drought tolerance, excellent graz...
